摘  要:目的探讨灯盏花素对糖尿病大鼠肝脏转化生长因子β1(TGF-β1)表达的影响。方法建立链脲佐菌素(STZ)诱导的糖尿病模型,应用Masson染色对肝组织作病理检查,Western印迹检测肝组织TGF-β1表达。结果灯盏花素给药组大鼠肝组织甘油三脂、胆固醇及游离脂肪酸水平低于模型组(P<0·05)。Masson染色显示模型组大鼠肝组织纤维化评分高于对照组(P<0·01);灯盏花素给药组大鼠肝组织纤维化评分低于模型组(P<0·05)。Western印迹显示模型组肝组织TGF-β1蛋白表达较对照组增加2·9倍,灯盏花素给药8周可使肝组织TGF-β1蛋白表达下降46%。结论灯盏花素对糖尿病大鼠肝组织纤维化抑制作用机制可能部分与下调TGF-β1表达有关。Objective To investigate the effect of breviscapine on the expression of transforming growth factor-β1 (TGF-β1 ) in liver in diabetic rats. Methods Diabetes was induced by injection of STZ. Liver lesion was evaluated using Masson's trichrome staining, and the expression of TGFβ1 was determined by Western blot analysis in liver. Results Levels of triglycerides, cholesterol and free fatty acids in liver in breviscapine - treated rats was markedly lower than that in diabetic rats. Masson's trichrome staining showed a significant increase in the amount of fibrosis in diabetic liver, liver fibrosis was significanly attenuated by breviscapine treatment. Western blot analysis showed a 2.9 fold increase in the amount of TGFβ1 in liver from diabetic rats with respect to control rats, breviscapine treatment could significantly reduced TGF-β1 protein expression by approximately 46%. Conclusion Brevescapine could inhibit liver fibrosis in diabetic rats, and its mechanism may be at least partly correlated with suppression on increased expression of TGF-β1 in liver.
