摘　　要：目的:检测大肠癌组织中Survivin基因的表达及凋亡指数,探讨二者的相关性及其临床意义。方法:采用分子信标荧光探针实时定量PCR法,检测49例大肠癌和14例大肠正常组织中Survivin基因mRNA模板拷贝数,TUNEL法检测凋亡细胞,计算凋亡指数(AI)。结果:肿瘤组织与正常组织比较Survivin基因模板拷贝数和AI差异均有统计学意义;肿瘤组织中mRNA拷贝数与肿瘤部位、性别、年龄和Dukes分期无关,与淋巴结转移和病理类型有关;结肠组AI显著高于直肠组;肿瘤组织中Survivin基因表达阴性组AI显著高于阳性组。结论:Survivin基因在肠癌组织中高表达,可成为预测淋巴结转移的参考指标。Objective： To investigate the correlation between transcriptional level of survivin gene and apoptosis index in colorectal carcinoma. Methods： The expression of survivin was detected by real time polymerase chain reaction （PCR） with molecular beacon probe in 49 cases of colorectal carcinoma and 14 cutting edge colorectal tissues. Apoptosis cells were screened by TdT-mediated dUTP-biotin nick end labeling （TUNEL） technique, and the apoptotic index （AI） was calculated. Results： The copies of survivin gene expression in colorectal carcinoma were significantly higher and apoptotic index in colorectal carcinoma was significantly lower （P〈0.05）. There were no correlation between the copies of survivin and tumor location, gender, age and Dukes stage of the patients. The copies of survivin gene expression were related to different pathological types and lymph node metastases. Apoptotic index was significantly higher in colons than that in rectums and lower in survivin expression positive samples than that in negative group, respectively. Conclusion： Survivin is overexpressed in colorectal carcinoma and may be an indicator in predicting lymph nodes metastasis.
