摘 要:目的探讨活血化瘀方对糖尿病大鼠模型创面肉芽组织不同时段(3、11d)转化生长因子-β(TGF-β)中TGF-β1 mRNA和TGF-β3mRNA表达水平影响,为"祛瘀利于生肌"的愈创理论提供科学依据。方法36只SD大鼠分别造成糖尿病溃疡模型和正常创面模型,随机分为活血化瘀组和糖尿病创面生理盐水对照组和正常创面生理盐水对照组。分别于创面修复3、11d取材,采用real-time PCR法检测新生肉芽组织中TGF-β1 mRNA和TGF-β3mRNA水平变化。结果创伤后3d和11d,中药活血化瘀组TGF-β1mRNA和TGF-β3 mRNA拷贝数均高于糖尿病创面生理盐水组(P<0.05),但低于正常创面生理盐水组(P<0.05)。结论活血化瘀方能在创伤后明显上调糖尿病大鼠模型新生肉芽组织中TGF-β1 mRNA和TGF-β3 mRNA表达,有利于创面愈合,证实了"祛瘀利于生肌"的愈创理论。Objective To investigate the influence of Huoxuehuayu Formula on the mRNA expressions of transform growth factor-β1 (TGF-β1) and TGF-β3 of new granulation tissue in different periods (3 days to 11 days) in diabetic rat model, and to provide a scientific base for TCM theory of "removing stasis being good for promoting regeneration". Methods The diabetic ulcer model and general wound model were established respectively in 36 SD rats, which then were randomly divided into the Huoxuehuayu Formula group, diabetic wound control group treated with normal saline solution (NSS) and normal wound control group treated with NSS. The samples were taken respectively 3 days or 11 days after the wound healing. The changes of mRNA expressions of TGF-β1 and TGF-β3 in new granulation tissue were detected by real-time PCR method. Results After 3 or 11 days in the Huoxuehuayu Formula group the copies of mRNA of TGF β1 and TGF-β3 were higher than those in the diabetic wound control group ( P 〈 0.05 ) but lower than those in the normal wound control group (P 〈 0.05 ). Conclusion Huoxuehuayu Formula can up-regulate obviously the expressions of mRNA of TGF-β1 and TGF-β3 mRNA of new granulation tissue in diabetic rat model, it is beneficial to wound healing and verification of the theory of "removing stasis being good for promoting regeneration".
