摘 要:目的以伞形科蒿本属植物川芎Ligusticum chuanxiongHort.叶为材料,研究川芎DNA的提取方法。方法分别采取CTAB法、高盐低pH法、木本植物DNA提取法、改良高盐低pH法提取基因组DNA,并对4种方法进行了改进。通过0.9%琼脂糖凝胶电泳和RAPD两种方法检测所提取的DNA样品。结果比较DNA产量、质量等,确定了木本植物DNA提取法最佳。结论木本植物DNA提取法为最佳方法。OBJECTIVE To study the medium for extraction of total Ligusticum chuanxiong DNA. METHODS The CTAB (cetyltrimethyle ammonium bromide) methods,Low pH extraction medium with high salt, the genomic DNA extraction method from the wooden plant, and Modified low pH extraction medium with high salt, were used to extract genomic DNA from the leaves of L chuanxiong. The DNA samples obtained by the methods above were tested by 0. 9% agarose gel electropboresis and PCR. RESULTS The difference in yield, quality of the DNA samples from the four methods were compared. CONCLUSION Extracted genomic DNA from the wooden plant medium is considered to be a fast, inexpensive and reliable technique among the four.
