摘 要:目的观察重组人生长激素(rhGH)对败血症大鼠炎症调控网络的影响。方法采用ipE.coli的方法复制大鼠败血症模型。42只♀SD大鼠随机分为正常对照组(C组)、败血症组(S组)及rhGH治疗组(T组)。S、T组又依观察时点再分为1d、3 d两个亚组。观测血浆肿瘤坏死因子α(TNFα)、白细胞介素-10(IL-10)、一氧化氮(NO)水平及诱生型一氧化氮合酶(iNOS)的表达情况。结果S1d组及T1d组血浆TNFα浓度均明显高于C组水平;S3d组血浆TNFα浓度接近C组水平,T3d组血浆TNFα浓度明显低于C组水平。S组血浆IL-10水平在各时点均显著低于C、T组水平,T组血浆IL-10水平在各时点与C组相比均无明显差异。S组血浆NO浓度及iNOS活力明显高于C组水平及T组水平,T组血浆NO浓度及iNOS活力仅于1 d时,明显高于C组水平。结论rhGH在减少败血症大鼠血浆TNFα、NO等促炎介质生成与释放的同时,也能有效地维持IL-10的抗炎介质水平,提示rhGH治疗可能有助于维持败血症时炎症调控网络的平衡。OBJECTIVE To investigate the effects of recombinant human growth hormone(rhGH) on inflammatory cytokine network of rat sepsis. METHODS E. coli was injected intraperitoneally to produce sepsis models. 42 female SD rats were randomly divided into three groups: control group (group C) , sepsis group( group S) and treated group( group T). Group S and group T were further divided into 1 d and 3 d subgroups. Levels of plasma tumor necrosis factor α (TNFα), interleukine - 10 (IL - 10), nitric oxide(NO) and activities of inducible nitric oxide synthase(iNOS) were determined. RESULTS On the 1 st day, plasma TNFα elevated significandy in group S and group T. On the 3rd day, TNFct in group S returned to the level of group C, while TNFct in group T went down below the level of group C. Levels of plasma IL - 10 in group S were obviously lower compared with that in group C and group T. Plasma IL - 10 in group T showed no significant difference than that in group C. Plasma NO and iNOS activities in group S elevated obviously than those in group C and group T. In group T, the level of NO and iNOS activities just increased significantly than those in group C on the 1 st day. CONCLUSION rhGH could not only inhibit the production and release of inflammatory mediators such as TNFct and NO, but also maintain the level of anti -inflammatory mediators such as IL- 10, which may suggest that rhGH treatment could help to keep the balance of inflammatory cytokine network in sepsis.
