摘 要:目的:探讨共移植体所构成的微环境对神经干细胞移植后向神经元分化的影响。方法:48只同一基因背景的Wistar大鼠,随机分为:NSCs移植组,共移植体移植组。在新生的同一基因背景鼠海马取神经干细胞和大脑皮质取血管内皮细胞,把单独NSCs和共移植体移植入同一基因背景大鼠MCAO造模后24h缺血半暗带区,Brdu标记其中NSCs,利用免疫荧光双标于3d,7d,14d,60d分别观察NSCs向神经元分化情况。结果:在7d,14d,60d三个时间点神经干细胞向神经元分化中,共移植体组神经元分化率高于NSCs组,差别有统计学意义(P<0.05)。结论:共移植体对移植入MCAO模型大鼠的NSCs向神经元分化有促进作用。Objective: To explore the co-transplanted implant of neural stem cells effects on the differentiation of neural stem cells (NSCs) to nerons. Methods: Forty-eight rats with same genetic background were randomly divided into 2 groups :NSCs group,co-transplanted implant of NSCs group. In this experiment,we used neonatal rats with same gene to get ceils. The co-transplanted implant of NSCs was composed of the NSCs isolated from hippocampi and the vascular endothelial cells were isolated from cerebra cortex. We separatedly transplanted the NSCs alone and the co-transplanted implant of NSCs to the ischemic penumbra of MCAO rats. The NSCs were labeled with Brdu before transplantation. We observed the differentiation of NSCs by immunofluorescence staining at 3d, 7d, 14d, and 60d after transplantiation. Results : At 7d, 14d ,and 60d,the differentiation percent of the co-transplanted implant of NSCs group was higher than that of NSCs group,and statistical difference showed P 〈 0. 05 . Conclusion: The co-transplanted implant of NSCs improves the differentiation of transplanted NSCs to nerons.
