摘  要:从江苏省某健康鹅群分离并筛选出一株病毒,经鸡胚传代培养和血清学试验,证明该毒株为新城疫病毒。应用电镜技术、生物学特性检测对其进行鉴定,结果表明:该毒株MDT为146.4h,ICPI为0.375,参照判定标准,确定该毒株为NDV弱毒株。通过RT-PCR方法,扩增出该毒株F基因,并进行克隆和序列测定,其F蛋白裂解位点的序列为112G-K-Q-G-R-L117,符合典型的NDV弱毒株的特征,从基因和分子水平上进一步证明该毒株为NDV弱毒株,通过绘制的系统发育进化树进行分析,表明该毒株为F基因Ⅰ型新城疫病毒,并将该毒株暂命名为G0405。A certain virus was isolated in Jiangsu province from a healthy goose flock. Hemagglutination (HA) tests, Hemagglutinin inhibition (HI) tests,electron microscopy and biological experiments were used to identify this virus, The results showed that the virus was Newcastle disease virus (NDV) and the mean death time(MDT)of chicken embryos was 146.4 h,the intracerebral pathogenicity index(ICPI) in 1-day-old chicken was 0.375. The numerical data were consistent with the standard of NDV avirulent strains. Fusion(F) gene of the virus strain was amplified by RT-PCR. The nucleotide sequence of F gene and the amino acids sequence that was encoded were analyzed and a phylogenetic tree was constructed based on obtained sequences of reference NDV strains from the domestic and other countries. The results revealed that the F gene belonged to genotype Ⅰ. Its cleavage site region of amino acid sequences of F protein was ^112G-K-Q-G-R-L^117,which was the characteristic motif of avirulent strains. The results further proved that the virus was a natural avirulent NDV,and then the virus was named with G0405.
