摘  要:从间接免疫荧光检测呈DVH阳性的病死鸭肝脏分离到3株(CD、HB、XC)病毒,电镜下病毒颗粒呈圆形、无囊膜,直径30~40nm,对氯仿、pH3.0、50℃,60min的处理不敏感,分离毒连续传代3次,全部鸭胚在24~60h死亡,胚体呈弥散性充血、出血,其致死鸭胚的能力能够被Ⅰ型DHV高免血清中和。分离毒人工感染1日龄雏鸭后出现与自然病例相同的临床症状和病理变化,并能重新分离到相同病毒。间接免疫组化检测自然病例和人工感染雏鸭的肝、脾、肾等均呈现DVH阳性。结果表明分离到的3株病毒均为Ⅰ型DHV,为有效预防DVH提供了有用的实验数据。Three virus were isolated from the liver of dead ducklings in outbreak and epidemic farms of Sichuan province,which had typical clinic symptom and pathological changes of duck virus hepatitis (DVH),and showed DVH positive using indirect fluorescent antibody (IFA). The virus was shown spherical,no envelope,30 to 40 nm in diameter by electron microscopy. The virus was resistant to chloroform,heat (50 ℃,60 min)and pH3.0. All duck embryos inoculated with the 3rd filial generation isolation virus were died in 24 to 60 h,and duck embryos were serious hyperemia and hemorrhage. When 10-day--old duck embryos inoculated with the mixture of DHV type I antiserum and the virus,no duck embryo dead.1-day-old ducklings experimentally infected with three strains of virus could arise the same clinical symptom and pathological changes as natural cases,and the virus could be isolated again. Liver,spleen,kidneys from dead ducklings infected with 3 strains of virus and natural cases,detected by indirect immunoperoxidase staining,were positive or strongly positive. These results demonstrate that the three strains were type I of duck hepatitis virus. This study could provide the useful data for effective preventing DVH.
