摘 要:目的以THP-1巨噬细胞源性泡沫细胞为研究对象,观察不同浓度黄芪多糖对细胞活力和凋亡以及对胆固醇流出的影响。方法将THP-1细胞诱导分化成泡沫细胞,用不同浓度黄芪多糖对其干预24h,XTY法检测细胞活力,Annexin V/PI染色、caspase3活性检测观察细胞凋亡情况。γ计数仪检测胆固醇流出。结果黄芪多糖呈剂量依赖性(10~100μg/ml)诱导THP-1巨噬细胞源性泡沫细胞凋亡,抑制细胞活力;促进胆固醇流出。结论黄芪多糖可影响THP-1巨噬细胞源性泡沫细胞活力,诱导凋亡,促进胆固醇流出。Objective To study the action of astragalus polysaccharides (Aps) on apoptosis , cell activity and cholesterol efflux in THP-1 macrophage-derived foam cell. Methods After exposure of the cultured THP-1 macrophage-derived foam cells to Aps at different dose for 24 hours, apoptosis of THP-1 macrophage-derived foam cells were determined by caspase3 , Armexin V/PI; cell activity were determined by XTT, cholesterol effluxl were determined by liquid scintillator, respectively. Results Apoptosis of THP-1 macrophage-derived foam cell was induced by Aps in dose-dependent manners. Cell activity was inhibited by Aps. Aps increased cholesterol efflux in THP-1 macrophage-derived foam cell with dose dependent pattern. Conclusion Aps may induce cell apoptosis and inbihits cell activity of THP-1 macrophage-derived foam cells ; Aps may increase cholesterol efflux of THP-1 macrophage-derived foam cells.
