摘 要:目的研究秦皮提取物对四氯化碳所致小鼠急性肝损伤的保护作用。方法60只雄性昆明小鼠随机分为正常对照组,模型对照组,阳性对照组,大?中?小剂量给药组。大?中?小剂量给药组分别按2.0,1.0,0.5 g/kg灌胃秦皮提取物;阳性对照组按0.1 g/kg灌胃水飞蓟宾;正常对照组和模型对照组以等体积生理盐水灌胃,共6 d。第5天,模型对照组、阳性对照组和给药组按2.0 m l/kg腹腔注射CC l4,正常对照组腹腔注射等体积生理盐水。比较各组丙氨酸氨基转移酶(alan ine transam inase,ALT)?超氧化物歧化酶(superoxide d ismutase,SOD)和丙二醛(m alond ialdehyde,MDA)含量。结果秦皮提取物能显著降低四氯化碳所致小鼠各组ALT含量的升高(P<0.05),同时能升高SOD的活性,降低MDA的含量(P<0.05)。结论秦皮提取物对四氯化碳所致小鼠急性肝损伤具有一定的保护作用。Objective To investigate the protective effect of ash bark extract against carbon tetrachloride inducing acute liver injury in mice. Methods Sixty male mice were randomly divided into normal control group, model control group, positive control group, high- dosage group, middle-dosage group and low-dosage group. The mice in the above groups were fed for 6 days. The drug groups were pretreated intragastrically with 2.0,1.0,0.5 g/kg ash bark extract, positive control group was pretreated intragastrically with 0.1 g/kg silybin. Control mice were given the same volume physiological saline. On the 5th day of feeding, the mice except normal control group were given intraperitoneally 2.0 mL/kg carbon tetrachloride. Twenty four hot, rs after the intoxication, the serum ALT, SOD activities and MDA content were measured in each group above. Results Ash bark extract significantly decreased carbon tetrachlor/de inducing acute hepatic injury with increasing the activity of ALT and the content of MDA, increased the activity of SOD in serum. Conclusion It is revealed that ash bark extract can prevent the CCl4 from inducing acute liver injury in mice through its antioxidation.
