摘  要:探讨阻断CD86协同刺激分子对自然流产模型孕鼠TGF-β1、MMP-9、TIMP-3和PAI-1的表达及妊娠结局的影响。实验组于妊娠第4.5天腹腔注射大鼠抗小鼠CD86单抗,实验对照组注射大鼠同型IgG2b,而正常妊娠组不作任何处理。于妊娠第13.5天计算胚胎吸收率;用免疫组化测定胎盘蜕膜组织中TGF-β1、MMP-9、TIMP-3和PAI-1的表达,并进行图像分析检测免疫组化染色灰度值(D)。结果:(1)实验组的胚胎吸收率显著低于实验对照组(x^2=7.441.P<0.05)(2)实验组母胎界面中TGF-β1和PAI-1的灰度值均显著低于实验对照组(P<0.05);实验组母胎界面中MMP-9的灰度值明显高于实验对照组(P<0.05);实验组母胎界面中TIMP-3的灰度值与实验对照组和正常妊娠组比较均无显著差异(P>0.05)。妊娠早期阻断CD86协同刺激分子可使TGF-∞1、MMP-9、TIMP-3和PAI-1分别通过各自不同的途径来发挥免疫耐受作用并且使自然流产模型孕鼠的胚胎吸收率降低至正常妊娠水平。The effect of blockage of costimulatory signal on the expressions of cytokine TGFβ, and matrix metalloproteinase-9 (MMP-9), tissue inhibitor of metalloproteinase-3(TIMP-3), plasminogen activator inhibitor-1 (PAI-1)and outcome of pregancy in abortion-prone model of mice was investigated by using CBA/J X DBA/2 matings as the abortion-model and CBA/J X BALB/c matings as the normal pregnancy model. This investigation was performed in three groups of mice, in which the abortion-prone models consisted of experimental and control experimental groups as well as the normal pregnancy group. In the experimental group and the control experimental group, 100 μg of rat anti-mouse monoclonal antibodiy against CD86 molecule in 200 μl PBS or the irrelevant isotype matched rat IgG2b were injected intraperitoneally to pregnant CBA/J mice on day 4. 5 of gestation, respectively. As for the normal pregnancy group, nothing was given to mice. On day 13. 5 of gestation, all the pregnant CBA/J mice were sacrified and the embryo resorption rates were calculated, and the immunohistochemical methods were used to detect the expressions of TGF-β, MMP-9, TIMP-3 and PAI-1 proteins~ It was demonstrated that the embryo resorption rate in the experimental group was significantly reduced in comparison with that in the control experimental group (x^2 =7. 441, P=0. 006), yet with no significant difference with that in the normal pregnant group (x^2 =0. 016, P〉0. 05).Meanwhile, compared with the control experimental group, the expressions of TGF-β and PAI-1 in the experimental group was significantly increased (P= 0. 010;P= 0. 003 respectively), also showing no signficant difference with the normal pregnancy group (P=0. 500).
