缺血预处理对猪移植肝脏保护作用的研究

摘　　要：目的通过分析小型版纳猪供肝经过缺血预处理后产生的保护效应,为缺血预处理的临床应用提供理论依据。方法将实验用小型版纳猪25只随机分成正常对照组(A组)、缺血再灌注组(I/R)、缺血预处理组(IP)。采用HSP70免疫印迹法检测HSP70的表达量,观察光镜下细胞形态学损害。结果IP组HSP70表达量明显增加(p<0.01),IP组的急性排斥反应和缺血再灌注损伤明显减轻;结论缺血预处理对移植肝产生了明显的保护效应,缺血预处理对HSP70的表达起正性调控作用。Objective To investigate the protective effect of the IP treatment on the liver function of the China miniature pig model, and provide theory for the clinical application of IP. Methods 25 （partially inbred） pigs were randomly divided into three treatment groups： normal group, I/R group and IP group. The biopsies from them were carried out to detect the expressions of HSP70 by immunoblot, Histopathology assessment to identify the cell morphology injury were performed by optical microscopy. Results There is no death during the operation. HSP70 expression is notably increasing （P〈0.01）and less injury of liver cellular microstructure happens in the IP group, Conclusion IP treatment can produce obvious protection effects on liver, And can generate positive regulation and control on the expression of shock protein.
