防喘合剂对哮喘豚鼠的作用机制研究

摘　　要：目的探讨防喘合剂对哮喘豚鼠的作用机制及量效关系。方法将80只豚鼠随机分为五组,其中2～5组豚鼠造模,然后每日上午各组分别给予蒸馏水、不同剂量防喘合剂、强的松灌胃,14d后观察引喘潜伏期、肺泡灌洗液(BALF)细胞组分、血清IL-4、INF-γ、TXB2、6-keto-PGF1α水平、血嗜酸性粒细胞(EC)和肥大细胞(MC)计数及胸腺、脾、肾上腺重量。结果各治疗组与模型组比较,均能延长豚鼠引喘潜伏期、降低BALF细胞总数和EC数、降低血EC和MC数、降低血清IL-4和TXB2水平、升高INF-γ和6-keto-PGF1α水平;各治疗组比较,除血EC和MC数、升高INF-γ水平三项指标无差异,其余指标合剂大剂量组作用优于小剂量组,呈剂量正相关,而合剂大剂量组与西药组作用相当;对照组、模型组、合剂大剂量组胸腺、脾及肾上腺重量比较无显著性差异,但西药组低于此三组,差异有显著性。结论防喘合剂对豚鼠具有免疫调节作用,能控制气道变应性炎症,降低气道高反应性;改善气道局部及全身的循环状态;未见明显的激素样副作用。Objectives： To explore the mechanism and the close -effect relationship of Fangchuan Mixture（FM, 防喘合剂 ） on guinea- pigs with asthma. Methods： 80 guinea- pigs were divided randomly into 5 groups, the guinea -pigs in the 2, the 3, the 4 and the 5 group were made modle. Subsequently, each group grinea -pigs were fed partly distilled water, Preinisone and FM with different doses. After 14 days, recorded the time of appearing gasp after the last touch stimulus; toke count of the cell routine of BALF and EC, MC in blood; mensurated the Serum IL - 4, INF -γ, TXB2,6 - keto - PGF1 concentration; weighed out the thymus, spleen, adrenal gland. Results： The above - mentioned indexes of the treatment groups were better significantly than of the model group. Compared in treatment groups, the changes of the number of EC and MC; and the serum INF -γ concentration had no significantly difference; The other indexes of the high dose group were better than of the small dose group, while had no difference in the prednisone group. The average weight of thymus, spleen, adrenal gland comparison, of prednisone group were lighter than of the normal guinea - pigs group, the model group and the high close group, while there were no difference among the 3 groups. Conclusion： In asthma guinea - pigs, FM could recover immunity inbalance, lighten allergic inflammation of bronchium and hypersuseeptibility, inprove the body and bronchial circulation, and had no side - effect like glucocrticoids.
