苦参碱对人胃癌细胞NCI-N87体外的增殖抑制作用

摘　　要：目的探讨不同浓度苦参碱对体外培养的人胃癌细胞NCI-N87的增殖抑制作用。方法NCI-N87细胞经不同浓度苦参碱处理后,采用CCK-8法检测药物对肿瘤细胞的抑制作用,观察细胞形态学改变,检测细胞DNA分布。结果苦参碱浓度>3.0mg/ml,细胞毒作用较大;浓度在0.5～2.0mg/ml时,对肿瘤细胞的生长有抑制作用,且呈时间剂量依赖性;浓度在0.1mg/ml时,对肿瘤细胞生长几乎无抑制作用。苦参碱作用后可增加G1期细胞所占的百分比,阻止细胞进入S期。结论苦参碱对人胃癌细胞NCI-N87增殖有抑制作用,并能阻抑细胞周期的进程。Objective： To investigate the inhibition of different concentrations of matrine on Proliferation of human gastric cancer cell line NCI - N87 cultured in vitro. Methods： NCI - N87 cells were treated by matrine in different concentrations and variablc durations. Ceil growth and proliferation were observed by CCK - 8 assay. Morphological changs of cell were observed, and cell - DNA distribution was assayed. Results： When the drug concentration was higher than 3.0mg/ml, the major effect was cell mecrosis. Matrine in the concentrations from 0. 5mg/ml to 2.0mg/ ml could inhibit proliferation of NCI - N87 cells as reveled by CCK - 8 assay. Increase in the percentage of G1 phase cell was observed by flow cytometry. Conclusion： Matrine can inhibit the proliferation of human gastric cancer cell line NCI - N87 efficiently by affecting the cycle of cells.
