摘  要:目的:观察5-羟色胺转运体基因(5-HTTLPR)和G蛋白β3亚单位基因(GNβ3 C825T)多态性在中国抑郁症患者中的分布情况及特点,探讨抑郁症发生的遗传机制。方法:采用聚合酶链反应和限制性片段内切酶的方法检测了180例抑郁症患者(其中男性84例,女性96例)及156例健康人对照组的5-HTTLPR和GNβ3 C825T基因多态性。结果:5-HTTLPR和GNβ3 C825T基因多态性在两组人群中的分布差异均有显著性,抑郁症组的5-HTTLPR SS基因型及S等位基因频率和GNβ3 825TT基因型及T等位基因频率均高于对照组(P<0.05)。进一步联合基因型分析显示:同时具有5-HTTLPRS和GNβ3 825T等位基因者患抑郁症的相对危险度(OR=3.25,P=0.001)比单独具有5-HTTLPRS等位基因(OR=1.817,P=0.01)或GNβ3 825T等位基因者高(OR=2.214,P=0.001)。结论:5-HTTLPR和GNβ3 C825T基因可能均是抑郁症的易感基因,且显示这两个基因在抑郁症的发病中存在微效协同作用。Objective: To assess whether 5-HTTLPR and GNβ3 C825T gene polymorphisms are associated with depression and explore the genetic mechanism of the pathogenesis of depression. Methods: The genotypes were determined with polmerase chain reaction and allele-specific restriction enzyme analysis. Depression patients (n=180) and sex and age-matched controls (n=156) were included in this study. Results: The frequency of 5-HTTLPR SS genotype, S allele and GNβ3 825TT genotype, 825T allele were significantly higher in the depression group than in the controls (P〈 0.05). Combined genotype analysis showed that individuals with both 5-HTTLPR S and GNβ3 825T allele (OR=3.25 ,P=- 0.001 )had a higher risk than those with 5-HTTLPR S allele (OR=1.817,P=0.01) or GNβ3 825T allele (OR=2.214,P= 0.001 ) only for depression. Conclusion: The 5-HTTLPR and GNβ3 C825T polymorphisms are predisposing genes of depression. Our data also suggest a significant interaction between the 5-HTTLPR S allele and GNβ3 825T allele in depression.
