摘 要:目的观察中药“益肾清火”方对牙周炎大鼠牙龈组织中性激素受体基因表达的调节作用。方法以64只SD大鼠作为实验对象,分为空白对照组和牙周炎模型组,后者建模成功后又分为模型对照组、造模后灌服中药高剂量组和等效剂量组。空白对照组和模型对照组动物灌喂生理盐水,造模后灌服中药组动物则灌喂中药3个月。收集各组动物牙龈组织,采用实时PCR技术分别检测雌激素受体(ERα、ERβ)和雄激素受体(AR) mRNA的表达情况,并进行统计学分析。结果用药3月后,各组雄性动物ERα mRNA表达无显著差异;造模后灌服中药高剂量组雌性动物ERα mRNA表达显著高于空白对照组和模型对照组(P<0.01),而其与等效剂量组比较无显著差异(P>0.05)。各组组内ERα mRNA的表达在雌雄动物之间有显著性差异(P<0.01)。ERβ、AR mRNA在各组间和同组不同性别动物间的表达均无显著差异(P>0.05)。结论中药“益肾清火”方可以在一定程度上提高牙周炎大鼠牙龈组织中雌激素受体 mRNA的表达。Objective To evaluate the effect of Yishenqinghuo recipe on the gene expression of sex hormone receptors in rats with experimental periodontitis. Methods SD rats were selected as the study objectives and randomly divided into blank control group (without periodontitis model, fed with saline) , model control group (with periodontitis model, fed with saline) , high dosage model group (with periodontitis model, fed with doubling dosage of Yishenqinghuo) and equivalent dosage model group (with periodontitis model, fed with clinical equivalent dosage of Yishenqinghuo). After being gavaged with medicine/saline for 3 months, the gingival tissues were collected to analyze the gene expression of estrogen receptors (ERα and ERβ) and androgen receptor (AR) by real-time PCR method. All the results were subject to statistical analysis. Results Three months after treatment, no significant difference in the ERα mRNA expression was observed among the male rats in each group. The ERα mRNA expression of female rats in high dosage model group was significantly higher than that in blank control group and model control group ( P 〈 0.01 ) , while there was no significant difference between high dosage model group and equivalent dosage model group( P 〉 0. 05). There existed a significant difference in the ERct mRNA expression between male and female rats in each group (P 〈0.01 ). However, there were no significant differences in the expression of ERβ and AR mRNA whatever among different groups or within the same group(P 〉0.05). Conclusion In some degree, Yishenqinghuo recipe can up-regulate the gene expression of sex hormone receptors in gingival tissues of SD rats with experimental periodontitis.
