摘 要:目的研究甘油诱发小鼠急性肾损伤后肾小管再生的动态改变。方法肌肉注射甘油建立小鼠急性肾损伤模型,测定相关肾功能指标并观察肾组织形态学改变;运用Ki67单抗免疫组化染色法检测细胞再生动态改变。结果甘油注射后小鼠肾损伤主要表现为早期肾脏肿胀、肾指数增加,以及血清尿素氮和肌酐急剧升高;肾小管腔内出现大量蛋白性物质,小管上皮细胞变性、坏死、脱落。免疫组化检测显示,甘油注射后第4天,损伤的肾小管出现大量Ki67阳性的再生小管上皮细胞,约为正常的9倍,核分裂相多。结论肾小管损伤后,主要由残留小管上皮细胞分裂增殖进行修复,增生细胞在损伤后第4天达到最高值。Objective To study the dynamic changes of cell regeneration after glycerin-induced acute renal injury in mice. Methods The model of acute renal injury of mice was induced with glycerin injection. Functional and morphologic changes were evaluated with routine methods. Dynamic changes of cell regeneration were judged by Ki67 antibody immunohistochemical staining. Results At the early period after glycerin injection, the kidney swelled, renal index was increased, blood urea nitrogen and creatinine were significantly increased, renal tubules were filled with protein mass, renal tubule cells suffered with degeneration, necrosis and desquamation. Immunohistochemical staining revealed that 4 days after glycerin injection, cell regeneration in tubules emerged in large amout, which was 9 times as the normal condition. They were Ki67-positive cells with more karyokinesis phase. Conclusion Regenerated cells were mainly originated from residual uninjured tubular cells and arrived at the highest level at the fourth day after glycerin injection.
