摘  要:目的优化早期胚胎的免疫荧光染色方法并观察蛋白激酶C(PKCα)在小鼠着床前胚胎的表达和定位。方法用优化的免疫荧光化学法结合激光共聚焦显微术,观察小鼠早期胚胎2-细胞期到囊胚期PKCα的表达情况。结果小鼠早期胚胎自2-细胞期到囊胚期均有PKCα亚型的表达,且主要集中在早期胚胎卵裂球的核内。结论经优化后的免疫荧光染色可清晰地检测到胚胎内PKCα集中于细胞核内表达,且分裂间期的表达高于分裂期。Objective To optimize the immunofluoreseent staining protocol and to observe the expression and location of the protein kinase Cα (PKCα) in preimplantation mouse embryos.Methods We observed the expression of PKCα in the preimplantation mouse embryo from 2-cell stage to blastula stage by using modified immunofluorescent staining and laser confocal microscopy. Results PKCα was expressed in nuclei from 2-cell stage to blastula stage during the preimplantation development. Conclusion The expression of PKCα can be detected clearly when the modified protocol was employed. The PKCα is localized in the nuclei, and it shows higher expression level during interphase than in the dividing phase.
