摘 要:目的:观察玉米幼芽提取物对雌性衰老小鼠卵巢和子宫抗氧化损伤方面的作用。方法:实验于2006-04/06在西北农林科技大学生理实验室完成。实验分组:选取健康雌性昆明小鼠28只,随机分为4组,每组7只,分别为正常组,衰老组,玉米幼芽提取物30mg/kg组和60mg/kg组。实验干预:正常组每天注射生理盐水,其余3组每天注射D-半乳糖(200mg/kg),连续45d。注射的第11天开始正常组,衰老组每天灌服蒸馏水,玉米幼芽提取物30mg/kg组和60mg/kg组加服30mg/kg和60mg/kg玉米幼芽提取物。末次灌胃禁食,确定间情期后处死,迅速解剖取出卵巢和子宫,低温下匀浆组织。实验评估:考马斯亮蓝法测定总蛋白含量,羟氨法测定超氧化物歧化酶活性、TAB法测定谷胱甘肽过氧化物酶活性,比色法测定丙二醛含量。结果:28只小鼠均进入结果分析。①总蛋白含量:衰老组显著低于正常组[卵巢(24.27±3.40),(50.84±3.65)mg/L,(P<0.01);子宫(38.49±1.71),(47.47±3.43)mg/L,(P<0.01)],玉米幼芽提取物30mg/kg组和60mg/kg组显著高于衰老组[卵巢(38.68±4.51),(38.37±0.55),(24.27±3.40)mg/L,(P<0.01);子宫(41.98±0.92),(42.96±0.84),(38.49±1.71)mg/L,(P<0.01)]。②超氧化物歧化酶活性:衰老组显著低于正常组[卵巢(20.10±3.98),(23.27±1.88)mkat/g,(P<0.05);子宫(85.00±9.70),(113.21±14.34)mkat/g,(P<0.05)],玉米幼芽提取物30mg/kg组和60mg/kg组显著高于衰老组[卵巢(27.52±2.55),(24.17±1.63),(20.10±3.98)mkat/g,(P<0.01,P<0.05);子宫(105.85±7.20),(113.06±5.10),(85.00±9.70)mkat/g,(P<0.05)]。③谷胱甘肽过氧化物酶活性:衰老组显著低于正常组[卵巢(0.0900±0.0050),(0.1050±0.0067)mkat/g,(P<0.01);子宫(0.0300±0.0800),(0.0633±0.1584)mkat/g,(P<0.01)],玉米幼芽提取物30mg/kg组和60mg/kg组显著高于衰老组[卵巢(0.1217±0.0083),(0.1134±0.0067),(0.0900±0.0050)mkat/g,(P<0.01);子宫(0.0817±0.0200),(0.0683±0.0750),(0.0300±0.0800)mkat/g,(P<0.01)]。④丙二醛含量:衰老组显著高于�AIM: To observe the protective effect of the extracts of maize plumule on the ovary and uterus from oxidation in female aged mice. METHODS: The experiment was conducted in the physiological laboratory of Northwest Science and Technology University of Agriculture and Forestry from April to June 2006. Twenty-eight healthy female Kunming mice were selected and randomly divided into 4 groups with 7 animals in each group: normal control group, aged group, 30 mg/kg and 60 mg/kg extracts of maize plumule groups. Except the normal group treated with normal saline, the others were injected with 200 mg/kg D-galactose daily for successive 45 days. After 10 days, the normal control group and aged group were given distilled water everyday, and the last two groups were fed 30 mg/kg and 60 mg/kg maize plumule extracts. All mice were fasting after the last time administration, and killed to take out the ovary and uterus and put into tissue homogenate. The total protein content, superoxide dismutase (SOD) activity, glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) were measured by Coomassie brilliant blue, hydroxylamine, TAB and chromatometry methods, respectively. RESULTS: Twenty-eight mice were involved in the result analysis.①The total protein content of the aged group was lower than that of the normal control group [ovary (24.27±3.40), (50.84±3.65) mg/L, (P 〈 0.01); uterus (38.49±1.71), (47.47±3.43) mg/L, P 〈 0.01]; that of the30, and 60 mg/kg extract groups was higher than that of the aged group [ovary (38.68±4.51), (38.37±0.55), (24.27±3.40) mg/L, P 〈 0.01; uterus (41.98±0.92), (42.96±0.84), (38.49±1.71) mg/L, P 〈 0.01]. ②The activity of SOD in the aged group was lower significantly than that in the normal control group [ovary (20.10±3.98), (23.27±1.88) nkat/μg, P 〈 0.05; uterus (85.00±9.70), (113.21±14.34) nkat/μg, P 〈 0.05]; that of the 30, and 60 mg/kg extract groups was higher than that of the aged gr
