摘  要:目的:体外培养成骨细胞,观察巴戟天多糖和巴戟天水提物对成骨细胞活性的影响。方法:实验于2004-09/2005-08在福建省骨伤研究所骨伤分子生物学实验室完成。①提取24h内新生SD大鼠头盖骨成骨细胞,分别用50,100,200g/L巴戟天多糖及50,100,200g/L巴戟天水提物药物血清进行体外培养。对照组用含50,100,200g/L无药血清的DMEM培养液培养。培养72h后进行相关指标检测。②通过MTT方法检测巴戟天多糖、巴戟天水提取物对成骨细胞增殖的影响。③检测碱性磷酸酶、细胞内骨钙素、转化生长因子β1基因表达等指标,观察巴戟天多糖、巴戟天水提取物对成骨细胞活性的影响。结果:①药物血清对成骨细胞增殖的影响:100g/L巴戟天水提物组和100g/L巴戟天多糖组吸光度大于对照组,差异有非常显著性(P<0.01),且100g/L巴戟天多糖组吸光度高于100g/L巴戟天水提物组(0.4302±0.0193,0.3829±0.0228,P<0.01)。②药物血清对成骨细胞活性的影响:巴戟天水提物组与巴戟天多糖组碱性磷酸酶比活性、细胞内骨钙素含量、转化生长因子β1mRNA相对表达量均高于对照组[碱性磷酸酶比活性:(16.89±2.09),(20.68±2.44),(11.23±2.40)μkat/g;骨钙素含量:(3.74±0.82),(3.97±0.69),(2.63±0.34)ng/106;转化生长因子β1 mRNA相对表达量:2.97±0.37,4.23±0.30,0.07±0.27,P均<0.01]。巴戟天多糖碱性磷酸酶比活性、转化生长因子β1 mRNA相对表达量高于巴戟天多糖组(P<0.01)。结论:巴戟天水提物与巴戟天多糖均能显著促进体外培养成骨细胞增殖、促进成骨细胞分泌碱性磷酸酶与骨钙素、促进成骨细胞转化生长因子β1 mRNA的表达,并且巴戟天多糖优于巴戟天水提物。AIM: To observe the effect of morinda root polysaccharide and its aqueous extract on the activity of osteoblast cultured in vitro. METHODS: The experiment was conducted in the molecular biological laboratory of Fujian Orthopaedics and Traumatology Institute from September 2004 to August 2005.①Osteoblasts were extracted from the parietal bone of SD rats bom in 24 hours, and cultured in drug serum of 50, 100, 200 g/L morinda root polysaccharide and 50, 100, 200 g/L aqueous extract, respectively. The control group was cultured in DMEM culture solution with 50, 100 and 200 g/L drug free serum. Seventy-two hours later, the related indexed were detected, ②The effects of morinda root polysaccharide and its aqueous extract on the proliferation of osteoblasts were detected by MTT.③The alkaline phosphatase lAP), osteocalcin (OC) and the expression of transforming growth factor β1 (TGF-β1) were also measured. RESULTS:① Effect of drug serum on osteoblast proliferation: The absorbances of the 100 g/L morinda root polysaccharide group and 100 g/L morinda root group were higher than that of the control group (P 〈 0,01), and that of the morinda root polysaccharide group was higher than the morinda root group (0.430 2±0.019 3, 0.382 9±0.022 8, P 〈 0.01 ), Effect of drug serum on the osteoblast activity: The AP, OC and expression of TGF-β1 of the morinda root polysaccharide group and morinda root group were higher than those of the control group lAP: (16.89±2.09), (20.68±2.44), (11.23±2.40) μkat/g; OC: (3.74±0.82), (3.97±0.69), (2.63±0.34) ng/10^th, TGF-β1 mRNA: (2.97±0.37, 4.23±0.30, 0.07+0.27), P 〈 0.01].
