摘  要:目的:研究单核细胞趋化蛋白-1(MCP-1)对人脐静脉内皮细胞(hUVEC)凋亡的影响.方法:培养并鉴定hUVEC;用不同浓度MCP-1(0.1,1.0,10,100μg/L)对其作用24,48h;Annexin V/PI双染细胞,流式细胞仪观察内皮细胞凋亡;琼脂糖凝胶电泳检测细胞染色体“DNA ladder”的形成.结果:MCP-1能诱导hUVEC的凋亡,其效应随浓度和时间的增加而增强(P<0.01);Annexin V/PI显示凋亡细胞明显增加;细胞DNA呈明显的“DNA ladder”.结论:MCP-1能诱导hUVEC凋亡.AIM: To study the effects of monocyte chemotactic protein-1 (MCP-1) on the apoptosis of human umbilical vein endothelial cells (hUVEC). METHODS: hUVEC were cultured in vitro and identified, hUVEC that growed vigorously were stimulated with different concentrations of MCP-1 (0.1, 1.0, 10, 100 μg /L)for 24, 48 hours. The early apoptotic events were detected with Annexin V/PI reagent kit by flow cytometry. Then the DNA fragments were observed by agarose gel electrophoresis. RESULTS: MCP-1 induced the apoptosis of hUVEC in a dosedependent manner and a time-dependent manner ( P 〈 0.01 ). Annexin- V/PI double staining assay showed that stimulation to hUVEC with different concentrations of MCP-1 enhanced the apoptosis rate (P 〈 0.01 ) as compared with that in control group. The characteristic pattern of DNA ladder was observed by agarose gel electrophoresis after 24-hour treatment of MCP-1. CONCLUSION: MCP-I can induce the apoptosis of hUVEC.
