摘 要:目的分析器官移植受者人巨细胞病毒(HCMV)临床分离株糖蛋白H基因型,为进一步研究其致病性、免疫性以及研制HCMV亚单位疫苗提供依据。方法以巢式聚合酶链反应(nested-PCR)检测97例器官移植受者外周血细胞中的巨细胞病毒gH基因并测序分型。结果有23例移植受者的标本经nPCR检出gH基因阳性,阳性率为23.71%,其中骨髓移植受者阳性率10.31%,肝移植受者阳性率5.15%,肾移植受者阳性率8.25%。在23例HCMV分离株中,20例得到gH基因型结果,其中gH1型10例,占50%,gH2型6例,占30%,gH1、2混合型4例,占20%,呈散在分布(χ2=4.00,P>0.05);gH基因型在各移植类型间也呈散在分布(χ2=5.652,P>0.05)。结论HCMV临床分离株糖蛋白基因分别属于gH基因型1~2型,呈散在分布。Objective To investigate genotypes of glycoprotein H of human cytomegalovirus(HCMV) clinical strains from transplant recipient in order to study it's pathogenicity, immunity and develop subvaccine. Methods Cytomegalovirus gH gene in blood samples of 97 bone marrow and solid organ transplant recipients were amplified by nested-PCR. Positive samples were directly sequenced to identify genotype. Results 23 (23.71%)HCMV gH amplificates were detected in strains of transplant recipients. Among them 10 strains(10.31%) were in bone marrow transplant recipients, 5 strains (5.15%) were in liver transplant recipients and 8 strains (8. 25%) were in kidney transplant recipients. 20 positive gH fragments were found in 23 amplificates. Among them 10 strains (50%) were gill genotypes, 6 strains (30%) were gH2 genotypes and 4 strains (20%) were gill and gH2 mixture type. No obvious difference was found between genotypes (χ2 =4.00, P〉0. 05) and type of transplantation ( 2 =5. 652,P〉0.05). Conclusion The gH 1-2 genotypes of human cytomegalovirus clinical strains were found scattered in transplant recipients.
