摘  要:目的对海洋放线菌WBF7进行菌种鉴定,并对其抗肿瘤代谢产物进行初步研究。方法通过形态学观察、培养特征和生理生化特性检测、细胞壁组分以及16S rDNA基因序列分析,鉴定菌株;将其发酵液pH调至2、7或10后100℃加热处理1h,研究活性物质的稳定性;用石油醚、乙酸乙酯及其水饱和正丁醇对其发酵液依次进行萃取,研究活性物质的极性;通过MTT法对处理后样品进行抗肿瘤活性研究。结果通过菌种鉴定,将菌株WBF7归属为灰略红链霉菌(Streptomyces griseorubens);其抗肿瘤活性物质具有较强的酸碱稳定性和热稳定性,并且易于被乙酸乙酯萃取。结论海洋放线菌WBF7是第一次从海洋中分离得到的灰略红链霉菌,其抗肿瘤代谢产物稳定性好,大部分极性中等,具有较好的开发前景。Objective To identify marine actinomycete WBF7 and study its antitumor metabolites. Methods The strain WBF7 was identified according to the 16S rDNA gene sequence analysis, the morphological, physiological and biochemical characteristics. Based on measuring antitumor activities by MTT assay, the pH stability, thermostability and polarity of antitumor metabolites were observed. Results Through strain identification, marine actinomycete WBF7 belonged to Streptomyces griseorubens. Its antitumor metabolites were very stable at pH 2410 and 100℃, and were mostly extracted by ethyl acetate, Conclusion It was the first report that S. griseorubens was isolated from the sea. The antitumor metabolites from the strain WBF7 were stable and moderate polarity.
