摘 要:目的观察丙烯腈(Acrylonitrile AN)所致的大鼠脑组织病理学改变及对神经特异蛋白质表达的影响。方法选用SD雄性大鼠,应用苏木素—伊红(HE染色法)染色、免疫组织化学、图像分析等技术综合评价经饮水染毒大鼠3个月后所致大鼠病理改变及对神经胶质纤维酸性蛋白(GFAP)和神经微丝(NF)的影响。结果低剂量染毒大鼠皮层和海马区胶质细胞增生,神经元排列紊乱,且高剂量染毒组还可见血管扩张和出血,但未见明显的神经元坏死迹象;免疫组织化学技术观察显示GFAP表达增加;NF表达减少,排列密集,且有串珠样改变。结论胶质细胞增生及NF改变为AN所致神经损害的病理学特征。Objective To evaluate neuropathologlcal characteristics and expression of nerve specific proteins in the male mt exposed to acrylonitrile(AN)in drinking water. Methods 30 male SD mrs were randomly divided into three groups, control group( n = 10),low dose group (n = 10), high dose group (n = 10), which were administrated 0, 50,200 mg/L AN in the drinking water respectively. The treatment was lasted for three months, Three animals were randomly selected from each group to be tested by H.E. staining and immuohistochemical methods for pathological evaluation. Results The degeneration of neurons and proliferation of astrocytes were observed in AN treated rats; and the expression of ghal fibriUary acidic protein (GFAP) increased with the ghosis in response to AN treatment, but the expression of neurofllament were decreased and disarranged. Conclusion Neurotoxic effects induced by AN may appear as the manifestation of molecular changes of GFAP, and the abnormality of NF, which may provide useful markers of neurotoxicity.
