肝癌特异性GGT-Ⅱ抗体制备及其电泳分析的临床应用

摘　　要：γ-谷氨酰移换酶同工酶II(GGT-Ⅱ)对肝癌诊断具有特异性.但其分析仅靠梯度聚丙烯酰胺凝胶电泳,方法繁琐,临床推广甚难.本研究将人肝癌血清中出现的GGT-Ⅱ酶蛋白提取并纯化,免疫动物制得抗人肝癌特异性GGT抗血清,建立免疫电泳分析方法,并用于肝癌的诊断.临床应用结果:对肝癌的诊断阳性率为86.9%,肝外肿瘤、孕妇、正常人全都阴性,而在急性肝炎、慢性肝炎和肝硬化标本中分别有15%、5%和10%表现为弱阳性.本文资料提示GGT-II的免疫电泳分析有助于肝癌的诊断,并具有开发推广价值.Although γ-glutamyl transferase isoenzyme II(GGT-II)was a specific marker for hepatoma diagnosis, thedetection of GGT-II was only separated by a polyacrylamide stage gel vertical electrophoresis, the procedure was moresteps and difficulty for clinical application. In this study, the enzyme proteins of GGT-II was abstracted and purified fromsera of patients.with pimary hepatic cancer(PHC) New Zealand rabbits was immunized using the purified enzyme protein asantigen.The rabbit antiserum against human GGT was used for GGT- II immunoelectrophoresis.The positive rate of GGT- II immunoassay was 86.9% in PHC, 15% in acute hepatitis, 5% in chronic hepatitis and 10% in liver cirrhosis respectively. Non rocket peak was found in sera of patients with non - liver tumors,gravidas and normal blood donors.The presentdata suggest that the immunoassay of GGT-II is a useful method for PHC diagnosis and easier to clinical application.
