摘 要:目的观察EOLA1在大鼠移植肝脏中的表达和功能。方法采用近交系雄性DA大鼠60只,Lewis大鼠120只,分为3组,改良“二袖套”法建立肝脏移植模型90例。A组Lewis为供体(n=30),Lewis为受体;B组DA为供体(n=30),Lewis为受体;C组以DA大鼠为供体(n=30),Lewis为受体,受体肝在移植后于门静脉注射RNA抑制性干扰质粒。观察平均存活时间;术后1、3、5、7、10时相点血清丙氨酸氨基转移酶(ALT)、总胆红素(TBIL)水平以及移植肝脏的病理学变化;半定量检测EOLA1蛋白的表达变化。结果平均存活时间A组超过100d,B组(16.2±1.4)d,C组平均存活时间(11.8±1.6)d;术后第5天B、C组血清ALT和TBIL升高较A组非常显著(P<0.01),B组高于C组差异显著;B、C组病理检查为明显的炎性反应,A组不明显;A组移植肝组织EOLA1表达最高,B组次之,C组最低,B、C组与A组比较差异非常显著(P<0.01),B、C两组间比较差异显著(P<0.05)。结论EOLA1蛋白表达水平的高低与肝组织内炎症反应的严重程度存在负相关;门静脉注射pSilencer质粒可以明显的抑制EOLA1在移植肝脏的表达。Objective To determine the changes in the expression of EOLA1 in the gratis under the interference of RNA through portal vein following DA→Lewis liver transplantafion(LT) and the function of EOLA1 in the grafts. Methods Ninety LT models were established by improved Kamada's two-cuff technique with inbred male DA rats( n = 60 )and Lewis rats( n = 30 )as the donors and inbred male Lewis rats as the recipients. The recipients were divided into three group A,B and C.A group was Lewis to Lewis and B group was DA to Lewis. C group was DA to Lewis following injection of RNA plasmid-interfering pSilencer through portal vein. Observed the average survival time of recipients. Detected the changes of serum biochemistry( Alanine aminotransferase ALT,Total bilimbin TBIL) and the grafts histology at day 1,3,5,7 and 10 after LT and the expression of EOLA1 protein. Results The average survival time of A group was over 100 days and B group was ( 16.2 ± 1.4) days but C group was ( 11.8± 1.6) days.The serum biochemistry and the grafts histology were significant changed after day 5 between A and B, C groups( P 〈 0.01 ). The changes between B and C was also significantly ( P 〈 0.05 ). The serum biochemistry of group C was higher and the grafts histology of group C was more sever than the results of group A and B. The expression of EOLA1 protein in the graft of group A was higher than group B and C( P 〈 0.01 ). The expression of EOLA1 protein in the graft of group B was also much higher than gmop C ( P 〈 0.05 ). Conclusion The level of EOLA1 expression is related to severity of acute infection of rejection of liver tissue after LT. We believe that they are negatively correlated. Injection of RNA plasmind-interfefing pSilencer through portal vein can certainly inhabit the expression of EOLA1 protein in the graft.
