摘 要:[目的]观察亚硒酸钠(Na2SeO3)、硫酸镁(MgSO4)对二氧化硅(石英)致巨噬细胞(RAW264.7)释放肿瘤坏死因子(TNF-α)、白介素-1(IL-1)的单独拮抗作用以及对人胚肺成纤维细胞羟脯氨酸(Hyp)合成水平变化的单独以及联合拮抗作用。[方法]对数生长期的RAW264.7细胞,调整浓度为2×10^5个/ml。在染200mg/L石英同时单独加入不同浓度Na2SeO3、MgSO4溶液,24h后测定培养液中TNF和IL-1水平,并取培养液与人胚肺成纤维细胞共同培养,48h后测定人胚肺成纤维细胞培养液中Hyp含量。另外,在染石英同时联合加入不同浓度的Na2SeO3和MgSO4溶液,培养24h后取培养液与人胚肺成纤维细胞共同培养,48h后测定其中的Hyp含量。[结果]①与石英组比,Na2SeO3对RAW264.7细胞释放TNF-α无明显影响,MgSO4可以显著降低TNF-α的活性(P〈0.05),其中以1.0μmol/L MgSO4的效果更为明显。(爹与石英组比,一定剂量的Na2SeO3、MgSO4可以降低IL-1的活性,其中以0.5μmol/L Na2SeO3和2.0μmol/L MgSO4的效果最好(P〈0.05)。③与石英组比,一定剂量的Na2SeO3、MgSO4可以显著降低人胚肺成纤维细胞Hyp合成水平(P〈0.05),其中以2.0μmol/L Na2SeO3和2.0μmol/L MgSO4的效果最明显。④与石英组比,一定剂量的Na2SeO3与MgSO4联合可以降低人胚肺成纤维细胞Hyp合成水平,其中以2.0μmol/L Na2SeO3和2.0μmol/L MgSO4联合作用效果最明显(P〈0.05)。[结论]一定剂量的MgSO4对石英致巨噬细胞释放TNF-α具有拮抗作用;一定剂量的Na2SeO3和MgSO4对石英致巨噬细胞释放IL-1具有单独拮抗作用,对人胚肺成纤维细胞Hyp合成水平具有单独及联合拮抗作用。[ Objective ] To investigate the effect of sodium selenite and magnesium sulfate which were used separately on TNF-α and IL-1 releasing from RAW264.7 macrophage treated with quartz and which were used separately and/or combined on the hydroxyproline synthetizing in human embryonic lung fibroblast also treated with quartz. [ Methods ] RAW264.7 and human embryonic lung fibroblast were cultured at 37 ℃ with 5% carbon dioxide. RAW264.7 were prepared during the growth period of logarithm in the concentration of 2 ×10^5/ml and poisoned with quartz( 200 mg/L ), at the same time sodium selenite and magnesium sulfate in different concentration were added separately. After 24 hours, TNF-α and IL-1 in RAW264.7 culture fluid were measured .Then the RAW264.7 culture fluid and the human embryonic lung fibroblast were cultured together. After 48 hours, the hydroxyproline in human embryonic lung fibroblast culture fluid were measured. In addition, when the RAW264.7 was poisoned with quartz, sodium selenite and magnesium sulfate were added together simultaneously. Then followed the same procedure as above mentioned and the hydroxyproline in human embryonic lung fibroblast culture fluid were measured too after 48 hours. [Results ] (1) Magnesium sulfate could reduce TNF-α releasing, with the middle dose ( 1.0 μmol/L ) most effective, but sodium selenite could not. (2)Sodium selenite and magnesium sulfate both could reduce IL-1 releasing, with the low dose ( 0.5 μmol/L )and high dose ( 2.0 μmol/L )most effective. (3)Sodium selenite and magnesium sulfate used separately could reduce the hydroxyproline synthetizing, with the high dose( 2.0 μmol/L )most effective. (4) Sodium selenite and magnesium sulfate used together can reduce hydroxyproline synthetizing, with both at the dose of 2.0 μmol/L most effective. [ Conclusion ] The specified dose of magnesium sulfate weakened the toxicity of quartz on TNF-α releasing in RAW264.7, but sodium selenite didn't. The specified dose of sodium sel
