摘  要:目的:观察绝经与骨质疏松的关系和绝经后以雌激素替代治疗的效果。方法:采取切除3月龄SD大鼠双侧卵巢方法,7wk后发现大鼠松质骨和密质骨骨量减少、骨小梁稀疏、骨皮质变薄、骨髓腔扩大等明显骨质疏松病理改变,同时伴有体重增加、肾上腺和子宫萎缩、血磷下降等现象。骨细胞水平研究表明去势后大鼠骨吸收增强,骨形成减弱,骨代谢处于负平衡状态。去卵巢同时灌胃给予30μg·kg-1·d-1雌激素(己烯雌酚)进行替代治疗。结果:去卵巢大鼠的骨量(尤其是密质骨骨量)丢失明显减少,骨组织显微结构病变和临床表现有较大改善。结论:雌激素替代治疗具有较好的防治骨质疏松效果。Objective\ To determine the relationship between ovariectomy and postmenopausalosteoporosis and to evaluate the effects of estrogen treatment in female rats. Methods\ Female Sprague Dawleg rats were subjected to control group (sham surgery), model group (OVX), and treatment group (OVX+30 μg·kg -1 ·d -1 estrostiben, ig). All rats were killed 7 weeks after surgery, their mainly organs were weighed, their blood Ca, P, ALP, ALT, estradiol, and testosterone tested, and their proximal and middle tibiae processed undecalcified for quantitative bone histomorphometry. Results\ Compared to the control rats, the trabecular and compact bone volume of model rats was markedly reduced; the bone tissue microstructure was obviously changed, such as trabecular thickness and number decreased, trabecular separation and medulla ossium cavity enlarged; the animal was accompanied by body weight increasing, adrenal gland uterusatrophy, and the blood concentration of phisphorus decreasing. The data of study in the bone cells indicated that the bone loss in model rats was associated with increasing in the bone absorption and decreasing in the bone formation. In contrast, treatment of OVX rats with 30 μg·kg -1 ·d -1 estrostilben resulted in normalization of trabecular and compact bone volume and improvement of pathological changes, the mechanism involved a decline in histomorphometric indices of bone resorption and an increase of bone formation. Conclusions\ Our results indicate that 7 weeks after bilateral ovariectomy in rats is able to establish an animal model of osteoporsis and estrogen treatment provides complete against osteopenia in OVX rats.
