体外阻断GR功能对鸡脾淋巴细胞活性及Dex免疫抑制的影响

摘　　要：用地塞米松（Ｄｅｘ）和ＲＵ４８６分别或联合与鸡淋巴细胞、ＣｏｎＡ一起培养，检测了淋巴细胞掺入３Ｈ－胸苷（３Ｈ－ＴｄＲ）量的变化。１０－５和１０－６ｍｏｌ／ＬＲＵ４８６可抑制淋巴细胞掺入３Ｈ－ＴｄＲ（Ｐ＜０．０１），而１０－７ｍｏｌ／Ｌ则无明显影响（Ｐ＞０．０５）；１０－５、１０－６和１０－７ｍｏｌ／ＬＤｅｘ对淋巴细胞掺入３Ｈ－ＴｄＲ有明显的抑制作用（Ｐ＜０．０１）；１０－５ｍｏｌ／ＬＲＵ４８６对Ｄｅｘ（１０－５～１０－７ｍｏｌ／Ｌ）的淋巴细胞掺入３Ｈ－ＴｄＲ抑制无明显逆转作用（Ｐ＞０．０５），而１０－７ｍｏｌ／ＬＲＵ４８６对Ｄｅｘ（１０－５～１０－７ｍｏｌ／Ｌ）的淋巴细胞掺入３Ｈ－ＴｄＲ抑制则有明显逆转作用（Ｐ＜０．０１）。将Ｄｅｘ、ＲＵ４８６分别与被ＣｏｎＡ活化２４ｈ的鸡淋巴细胞一起培养，１０－５ｍｏｌ／ＬＲＵ４８６可抑制淋巴细胞掺入３Ｈ－ＴｄＲ（Ｐ＜０．０１），而１０－６～１０－７ｍｏｌ／Ｌ则无影响（Ｐ＞０．０５）；１０－５～１０－６ｍｏｌ／ＬＤｅｘ可抑制淋巴细胞掺入３Ｈ－ＴｄＲ（Ｐ＜０．０５），１０－７无影响（Ｐ＞０．０５）。结果表明，高浓度的ＲＵ４８６（１０－５ｍｏｌ／Ｌ）可阻断糖皮质激素受体（ＧＲ?The effects of RU486,Dex alone and combined on the Con A stimulating responses of lymphocytes from chicken spleen were studied in vitro. Dex suppressed lymphocytes activity at 10 -5 , 10 -6 , 10 -7 mol/L, RU486 suppressed lymphocytes activity at 10 -5 , 10 -6 mol/L,but not at 10 -7 mol/L. There was no reversal of Dex induced suppression of lymphocytes ConA stimulating activity when lymphocytes were cultured with RU486 10 -5 mol/L mixed with Dex 10 -5 ￣10 -7 mol/L seperately and further suppression was not found. Dex induced immunosuppression was significantly reversed when lymphocytes were cultured with RU486 10 -7 mol/L mixed with Dex 10 -5 ￣10 -7 mol/L and the effect was Dex dosage dependent. When the concentrations of Dex increased,the reverse effect of RU486 10 -7 mol/L decreased. This indicated that Dex induced immunosuppression was mainly mediated via a GR mechanism and the immunosuppression could be reversed by blocking GR with RU486 at a suitable concentration. When the lymphocytes were activited by ConA 10 μg/ml for 24 hours,the Dex induced immunosuppression was found at 10 -5 and 10 -6 mol/L,RU486 induced immunosuppression was found at 10 -5 mol/L only. The results show that the sensitivity of lymphocytes to RU486 and Dex reduced when activitied by ConA for 24 hours.
