含日本脑炎病毒pre-M/E蛋白基因真核表达质粒的构建及免疫效果观察

摘　　要：本文作者将日本脑炎病毒（ＪＥＶ）的前膜蛋白和囊膜蛋白（ｐｒｅＭ／Ｅ）的基因插入到真核表达载体ｐＳＧ５中，构建了重组表达载体ｐＳＧＪＥ。用其体外转染地鼠肾细胞ＢＨＫ－２１，经间接免疫荧光法（ＩＦＡ）检测，证实了ＪＥＶ－Ｅ的瞬时表达。大量制备和纯化重组质粒，经肌肉注射免疫ＢＡＬＢ／ｃ小鼠，结果显示该重组质粒可明显促进小鼠在受到ＪＥＶ活病毒攻击后特异性抗体的产生。We inserted JEV preM/E gene into the eucaryotic expression plasmid pSG5. The recombinant plasmid, designated as pSGJE,was identified by in vitro transfection of BHK 21 cells. The transient expression of JEV E in BHK 21 cells was detected by indirect immunofluorescence assay(IFA) with a monoclonal antibody 2H4 against JEV E. Then we immunized BALB/c mice with purified pSGJE by intramuscular injection and the result showed that this recombinant plasmid could stimulate specific antibody response against JEV in mice.
