摘 要:目的探讨卡维地洛对柯萨奇病毒B3(CVB3)病毒性心肌炎小鼠的作用。方法随机将80只小鼠分为3组:正常对照组(n=20),心肌炎组(n=30)与卡维地洛组(n=30),利用BALB/c小鼠感染CVB3建立病毒性心肌炎模型,感染24 h后每日灌胃给予卡维地洛10 mg/kg直至第14天末,分别于接种第7天和第14天随机从各组抽取若干只小鼠取血后处死并留取心脏等标本。结果卡维地洛降低病毒性心肌炎小鼠急性期的病死率,显著减轻心脏病理损伤,显著减低HW/BW(HW,心脏重量;BW,身体重量)比值;卡维地洛干预后第7天小鼠心肌IFN-γ表达水平显著高于心肌炎对照组,MDA含量显著降低;干预后第14天SOD含量显著增加,MDA含量显著降低。结论卡维地洛通过增加心肌IFN-γ表达水平与抗氧化的双重作用减轻感染小鼠的心肌损害,提高生存率,起到保护性作用。Objective To examine the effects of carvedilol in a murine model of viral myocarditis indueod by coxsackievirus B3 infection(CVB3). Methods BALB/c mice were intraperitoneally inoculated with CVB3, 24 h after infection carvedilol ( 10 mg/kg) was orally administered for 7 or 14 consecutive days. Infection group and normal group were used with same dose PBS. Mice were killed at day 7 and 14. The survival rate and myocardial histopathology changes were observed. The HW/BW ratio, the myocardial IFN-γ production, the content of malondialdehyde (MDA) and superoxide dismutase(SOD) were studied. Results Carvedilol improved the 14-day survival of the animals, attenuated myocardial lesions and decreased HW/BW ratio. On day 7, carvedilol group increased the myocardial IFN-γ production and decreased the content of MDA more significantly than virus infection group . On day 1 4 , in carvedilol group we found increased SOD and decreased MDA . Conclusion Carvedilol protects animals against CVB3-induced viral myocarditis. Carvedilol exerts some of its beneficial effects by boosting the production of IFN-γ and by inhibiting peroxidants in addition to improving cardiac function.
