摘  要:目的观察肝宁颗粒对α-萘异硫氰酸酯(ANIT)所致大鼠急性黄疸模型的退黄降酶作用。方法将Wistar大鼠随机分为正常组、模型组、肝宁颗粒高、中、低剂量组和茵栀黄注射液组。各治疗组连续灌胃给药7 d后,ANIT诱发黄疸模型,48h后摘眼球取血,分离血清,以血清肝功能指标、SOD的活力及MDA含量以及肝脏组织病理学的改变为观察指标。结果与模型组比较,肝宁颗粒高、中剂量组、茵栀黄组的ALT,AST,TBIL,ALP,GGT,MDA均显著下降(P<0.01或P<0.05),SOD显著升高(P<0.01或P<0.05);与茵栀黄注射液比较,肝宁颗粒各剂量组的TBIL均显著增高(P<0.01),肝宁颗粒高剂量组的ALT,GGT,SOD,MDA均无差异(P>0.05)。肝脏病理组织学检查表明,肝宁颗粒能明显减轻肝细胞变性、坏死和肝小胆管增生。肝宁颗粒高、中、低剂量组之间存在量效关系。结论肝宁颗粒剂具有降低实验性胆汁淤积大鼠血清胆红素、转氨酶和改善肝脏组织损伤的作用,疗效略低于茵栀黄注射液。OBJECTIVE To observe the action of ganning granula on reduced jaundice and enzyme in jaundice model of rat induced by ANIT. METHODS Wistar rats were randomized into six groups: group A in which no treatment was given, group B in which ANIT was given on the seventh day, group C in which high dosage of "Ganning granula" was given, group D in which moderate dosage of "Ganning granula" was given, group E in which low dosage of" Ganning granula, was given, and group G in which "Yinzhihuang injection" was given as positive control. Except group A and B, the other groups were given corresponding medicines for 7 days and then additionally administered ANIT to induced jaundice. After 48 h, the rats'blood was gained from eyes. The serum was separated. The liver functional sign and the content of SOD and MDA in serum, and the change in pathology of liver tissue were observed. RESULTS Compared to group B, ALT, AST, TBIL, ALP, GGT, MDA of group C and D and G group dropped obviously ( P 〈 0.01 or P 〈 0.05) ; and SOD went up obviously ( P 〈 0. 01 or P 〉 0. 05). By the examination the liver pathology, we found that "Canning granula" could notably alleviate hepatocell change and necrosis and hepatominieholange hyperplasia. ALT, SOD, MDA and GGT of group C was not indifference with group G ( P 〉 0.05 ). CONCLUSION "Ganning granula" has the action of reducing serum bilirubin and transaminase and improving liver tissue injury in rats of experimental Cholestasis. But it' s thapeutie effect is not as good as "Yinzhlhuang injection".
