摘  要:目的探讨体外联合应用激活素A、全反式维甲酸(ATRA)、碱性成纤维细胞生长因子(bFGF)和烟酰胺4种生长因子能否将小鼠胚胎干细胞(ESC)诱导分化为胰岛素生成细胞团。方法先后以激活素A、ATRA、bFGF和烟酰胺4种生长因子诱导小鼠ESC 14 d后,应用RT-PCR、DTZ染色和免疫荧光检测分化细胞胰岛素表达。结果分化细胞可检测到胰岛素mRNA表达,DTZ染色和免疫荧光染色阳性。结论体外联合应用激活素A、ATRA、bFGF和烟酰胺4种生长因子能够将小鼠ESC诱导分化为胰岛素生成细胞团。Objective To induce mouse embryonic stem cells (ESCs) to differentiate into insulin-producing cell clusters by activin A, all-trans-retinoic acid (ATRA) , basic fibroblast growth factor (bFGF) and nicotinamide in vitro. Methods Mouse ESCs were treated by activin A, ATRA, bFGF and nicotinamide for 14 d, and then insulin expression was examined by reverse transcripton-polymerase chain reaction (RT-PCR) , DTZ-staining and immunofluorescence. Results The mRNA for insulin was detected in the differentiated cells, and DTZ-staining and immunofluorescence staining were positive. Conclusion Mouse ESCs can be induced to differentiate into insulin-producing cell clusters by activin A, ATRA, bFGF and nicotinamide in vitro.
