摘 要:应用修饰的家蚕核型多角体病毒为载体在家蚕细胞和幼虫中表达日本血吸虫中国大陆株28KD胱甘肽S-转移酶基因。Southern杂交证实该基因被插入到病毒基因组中的正确位置。在家蚕培养细胞中的产量为0.77mg,在家蚕幼虫中则超过5mg/条蚕。The gene of the 28kD glutathione S-transferase(GST) from the Chinese strain of Schistosoma ja ponicum had been expressed in the silkworm (Bombyx mori ) cells and larvae by Bombyx mori nuclear polyhedrosis virus (BmNPV) vector which had been modified. The GST gene was inserted into the right position of the BmNPV genome as identified by Southern hybridization. The product was a 28kD protein, had GST activity and antigenicity. The yield in BmN cells(1×10~6 cells/ml) was 0.77mg and more than 5mg in a silkworm larvae. All the results showed that it is possible to develop the GST expressed in the silkworm larvae to a new kind of genetic engineered schistosomiasis vaccine.
