摘  要:目的进一步探讨新生大鼠卵巢中Foxo3a(forkhead box group O)转录因子参与卵母细胞凋亡的调节机制。方法取出生后1、2、3和4d龄大鼠的卵巢,用免疫组化方法观察Foxo3a、Bim和FasL在卵巢组织中的表达水平以及细胞定位,并且应用TdT介导的dUTP缺口末端标记(TUNEL)技术检测卵巢内卵母细胞的凋亡变化。结果免疫组化显示Bim和FasL都在新生大鼠部分卵母细胞巢和原始卵泡的卵母细胞核内和胞浆高表达,且阳性率曲线都在2d达到高峰,随后逐渐降低,与TUNEL和Foxo3a的阳性率变化曲线一致。然而Bim和FasL的阳性率都低于Foxo3a的阳性率。结论Foxo3a可能同时诱导Bim和FasL的表达从而调节卵母细胞的凋亡。[Objective] To further investigate the association of Foxo3a expression with oocytes apoptosis in neonatal rat and detect the pathways by which Foxo3a regulates oocyte apoptosis. [Methods] Ovaries were collected from the postnatal rats on day 1, 2, 3, and 4 after birth. The proteins expression of Foxo3a, Bim, and FasL were examined by immunohistochemistry. The ovary apoptotle profdes were determined by terminal deoxynueleotidyltrans- ferase-mediated dUTP nick-end labeling (TUNEL). [Results] The expression of Foxo3a, Bim,FasL in oocyte nests and primordial follicles were detected in the nuclei and cytoplasms of some ooeytes. The patterns of Bim and FasL peoitive rates were consistent with that of Foxo3a and TUNEL positive oocytes rates, which reached the peak in the ovaries of 2--day-old rats. HoweverHHh, the positive rates of Bim and FasL were somewhat lower than that of Foxo3a, respectively. [Conclusion] Foxo3a, in neonatal rat ovaries, probably play an effect on the apoptosis of follicular oocytes by inducing the expression of Bim and FasL.
