酶联免疫吸附试验法测定国产及日产乌苯美司影响粒细胞-巨噬细胞克隆刺激因子分泌量的比较

摘　　要：采用新的夹心酶联免疫吸附试验（ＥＬＩＳＡ），测定不同浓度乌苯美司（ｕｂｅｎｉｍｅｘ，Ｕｂｅ）与１×１０６／ｍｌ人外周血单核细胞共同培育下的克隆刺激因子（ＣＳＦ）水平。当加入０．１μｇ／ｍｌ国产Ｕｂｅ时，粒细胞－巨噬细胞克隆刺激因子（ＧＭ－ＣＳＦ）浓度为３０１±１７ｐｇ／ｍｌ，相同浓度的日产Ｕｂｅ，其ＧＭ－ＣＳＦ浓度为３０９±１８ｐｇ／ｍｌ，两者之间无显著差异（Ｐ＞０．０５）。结果表明，Ｕｂｅ体外对人外周血单核细胞分泌ＧＭ－ＣＳＦ分泌量也提高。The levels of colony stimulating factor (CSF) in culture supernatant of 1×10 6/ml peripheral blood mononuclear cells (PBMC) cocultured with various concentrations of ubenimex (Ube) were determined using newly developed sandwich enzyme linked immunosorbent assay (ELISA).The concentration of granulocyte/macrophage CSF(GM CSF) in the supernatant in the presence of 0.1μg/ml domestic Ube was 301±17pg/ml,while imported Ube was 309±18pg/ml( P > 0.05 ).These results indicated that Ube enhanced on production of GM CSF from PBMC in vitro.And the production of GM CSF increased with increasing amounts of added Ube.The action of domestic Ube was as same as that of imported Ube.
