摘 要:目的观察4种常用的保存羊膜在不同保存期组织形态和超微结构的改变,为临床应用提供依据。方法取健康剖宫产产妇胎盘羊膜,随机分成4组保存:DMEM/甘油深低温组、纯甘油4℃组、无水酒精组、真空干燥组。于保存后1个月、3个月、12个月(DMEM/甘油组和纯甘油组),行HE染色光镜及透射电镜检查,观察其组织形态结构变化,以新鲜羊膜作为对照。结果(1)DMEM/甘油深低温保存组:光镜示保存1个月、3个月时羊膜组织结构基本完整,12个月时上皮细胞轻度破坏;透射电镜示在保存的各个时段羊膜上皮细胞完整,胞核局部溶解,线粒体肿胀,基底膜正常;(2)纯甘油4℃保存组:光镜观察保存1个月、3个月、12个月羊膜上皮细胞结构破坏,12个月基质层结构轻度破坏;透射电镜示保存各时段羊膜上皮细胞破坏明显,但基底膜基本正常;(3)无水酒精保存组:光镜示保存1个月、3个月时羊膜组织结构较完整;透射电镜示羊膜上皮细胞胞膜破坏,胞质、胞核溶解,基底膜变薄;(4)真空干燥保存组:光镜示保存1个月、3个月羊膜上皮细胞和部分基质层结构破坏明显;透射电镜示1个月、3个月时羊膜上皮细胞胞膜完整,核均质化与溶解并存,基底膜变薄。结论4种方法中,就组织形态和超微结构而言,以DMEM/甘油深低温保存法保存效果最好,纯甘油保存法次之,无水酒精和干燥保存法差。Objective To observe the morphology and ultrastructure changes of four preserved types of amniotic membranes (AM) during different periods,and provide the evidences for clinical application. Methods The AM from the healthy puerpem were randomly divided into four groups: DMEM/glycerol preserved group at - 80℃, glycerol preserved group at 4℃, ethanol preserved group and vacuum dry storage group. After preserved for 1 month, 3 months and 12 months( DMEM/glycerol and glycerol group), all their morphology changes were observed by light microscope and transmission electron microscope (TEM) ,the fresh AM was set as control group. Results ( 1 ) DMEM/glycerol group:Light microscope showed,AM structures were basically intact after preserved for 1 month and 3 months,but a few epithelial cells were destroyed after 12 months;TEM showed, the structures of epithelial cells in AM preserved for different periods were intact,nucleus dissolved partly and mitochondria showed tumefaction,while basement membrane still was normal; ( 2 ) Glycerol group: Light microscope showed, the structure of epithelial cells was destroyed in AM preserved for 1 month,3 months and 12 months,and stromal structure was destroyed after 12 months;TEM showed,the epithelial cells were destroyed obviously,but basement membrane was basically normal; ( 3 ) Ethanol group: Light microscope showed,AM structure was kept well after 1 month and 3 months, but TEM showed the epithelial cell membrane was destroyed, cytoplasm and nucleus were dissolved, basement membrane became thin; ( 4 ) Vacuum dry group:The epithelial cells and several stromal structures were destroyed evidently after 1 month and 3 months examined by light microscope, and the epithelial cell membrane was kept intact after 1 month and 3 months,but nucleus dissolved, and basement membrane became thin examined by TEM. Conclusion As for morphology and ultrastructttre,AM preserved in DMEM/glycerol can keep structures well,better than in glycerol,ethanol and dry stor
