摘 要:目的:利用彩色多普勒超声观察早幼粒细胞白血病蛋白(PML)基因对小鼠肝癌的抑瘤效果,探讨其应用价值。方法:18只裸鼠随机分为3组:①空白对照组,肝癌细胞未经处理;②重组腺病毒(Ad)-对照组,肝癌细胞经不含PML基因的重组Ad处理,为阴性对照;③Ad-PML组,肝癌细胞经重组Ad介导的PML基因处理。分别将上述各组肝癌细胞注射到裸鼠的皮下,每周观察肿瘤生长情况,6周后行超声检查。另将未经处理的肝癌细胞注射于12只裸鼠皮下,待肿瘤长出后随机分为2组,分别用Ad-PML和Ad-对照各0.1ml围绕肿瘤多点注射,2周后再重复注射,每周测量肿瘤大小并计算体积,6周后行超声检查。结果:①Ad-PML组裸鼠皮下未见肿瘤生长,而空白对照组及Ad-对照组裸鼠皮下可见肿瘤生长;超声观察两组肿瘤大小、回声及肿瘤体积之间差异无统计学意义(P〉0.05)。②周围注射Ad-PML的肿瘤较Ad-对照组明显小,二者之间差异有统计学意义(P〈0.05);超声显示二者之间内部回声及边界有差异。结论:重组Ad介导的PML基因能抑制肝癌的生长,超声可观察肿瘤生长的情况,为肿瘤抑制基因抑瘤效果的评价提供有价值的信息。Objective To observe the effect of recembinant adenoviral-mediated promyelocytic leukemia protein(PML) gene inhibition on liver cancer of mice with color Doppler ultrasonography(US) and to evaluate the application of US. Methods Eighteen nude mice were divided into three groups at random, the blank contrast group in which the HepG2 carcinoma cells were dealt with nothing, the adenovirus(Ad)-contrast group with the cells handled with adenovirus,the Ad-PML group with the cells transfected by the PML gene mediated by adenovirus. The HepG2 carcinoma cells of the three groups were injected into the subcutaneous tissue of the mice and the growth of the tumor were observed every week, and US were applied after 6 weeks. Furthermore,the carcinoma cells without disposal were injected into the subcutaneous tissue of 12 nude mice,and were divided into 2 groups at random after the tumor developed,then 0. 1 ml Ad-PML or Ad-contrast were injected at several dots circling the tumors,and the liquid above was injected a second time after 2 weeks. Then the parameters of tumor such as diameter and the volume were measured and calculated every week,and the US were applied after 6 weeks. Results ①There was no subcutaneous tumor in the Ad-PML group but there were tumors in the blank contrast group and Ad-contrast group. There were no obvious differences between the two groups in the diameter,echo and volume( P 〉0. 05). ②The tumors injected with Ad-PML were obviously smaller than the ones injected with Ad-contrast, and there was significant difference between them( P〈0.05). There was difference on the echo, border and the color Doppler flow between them. Conclusions PML gene mediated by recombinated adenovirus could inhibit the liver cancer growth. US could observe the tumor growth and provide the valuable information for the evaluation about the tumor inhibition.
