摘 要:目的:研究半枝莲的 HPLC 指纹图谱的测定方法,并确定其指纹图谱。方法:采用 Discovery-C_(18)柱(4.6 mm×250 mm,5 μm),以甲醇-水(0.05%甲酸)的溶剂系统线性梯度洗脱[0 min→30 min→70 min,甲醇-水(0.05%甲酸)比例为25:75→40:60→75:25],流速1 mL·min^(-1),检测波长335 nm,柱温24℃,进样量10 μL。结果:通过对10批不同产地半枝莲药材的研究建立了半枝莲药材的指纹图谱,其中13个色谱峰为共有峰。利用 HPLC 指纹图谱相似性评价软件评价,10批药材的相似度在0.985~0.744之间。结论:利用半枝莲指纹图谱可以对半枝莲药材进行质量评价。该方法的优点在于提取方法简单,用HPLC 即可使大部分化学成分得到较好的分离。数据表明,该法具有很好的稳定性、重复性和可行性。可为半枝莲药材提供更为科学的依据和有效的鉴别方法。Objective:To study the determination method of Banzhilian( Herba Scutellariae Barbatae) and to establish the fingerprint of Banzhilian. Methods:The methanol extraction of Banzhilian was separated on a Discovery - C 18 column (4. 6 mmo × 250 mm, 5 μm) eluted with CH3OH and H2O (0. 05 % CHOOH ) in linear gradient condition [ 0 min→30 min→70 min, elute rate of CH3OH and H2O (0. 05% CHOOH ) was 25 : 75→40 : 60→75 : 25 ] with DAD detected at 335 nm. Flow rate was 1 mL · min^- 1 and 10 μL was injected and the column temperature was 24 ℃. Results :The fingerprint of Banzhilian with 13 common peaks was established by study of 10 samples of different sources. Estimation the fingerprint by the similarity analysis of HPLC fingerprint spectra. The result of comparability was between 0. 985 - 0. 744. The similarity was good. Conclusions: The fingerprint can be used to distinguish Banzhilian medicinal materials. The advantage of this method is the simple extraction and the most chemical components can be separated very well by HPLC. This method has good stability repeatability and feasibility. Therefore it provides scientific reference and effective determining ways for the quality control of Banzhilian medicinal materials.
