摘 要:目的探讨一种可靠的肿瘤细胞分化模型,确立一套判定肿瘤细胞是否分化的简便、准确的方法。方法以分化诱导剂二甲基亚砜(dimethylsulphoxide,DMSO)影响下不同时间点的HL-60细胞为检测对象,应流式细胞术来分析细胞大小及细胞表面的分化标志物;经碘化丙啶(propidium iodide,PI)染色后,用激光共聚焦显微镜观察对已分化细胞进行形态学确认。结果随着药物诱导时间的延长,被诱导的HL-60细胞体积逐渐增大;在48h后,被DMSO诱导细胞开始表达分化标志物CD11b并出现细胞核型的变化。结论DMSO能诱导HL-60细胞分化;应流式细胞术来分析细胞大小及细胞表面的分化标志物,再用激光共聚焦显微镜观察已分化细胞核的形态变化是判定肿瘤细胞分化的简便、准确的方法。Objective To establish a constant model of malignant tumor cell differentiation and to set up a set of methods that the differentiated cells can be identified. Methods HL-60 cells influenced by dimethylsulphoxide (DMSO) capable of inducing differentiation for different time were used as subjects. We analysed the differentiation marker on the cell surface and cell volume by using flow cytometry. The differentiated cells have been identified by confocal microscope after having been stained with pro- pidium iodide (PI). Results With drug-inducing time increasing, the differentiated cells volume was enlarged gradually. After 48 hours, the differentiated cells induced by DMSO began to expressed differentiating marker CDI I b and the differentiated cells, nuclei was changed into the nucleus morphodifferentiation just like neutrophil nuclei. Conclusions The differentiation of HL-60 cells can be induced by DMSO. The cells volume and the cells differentiating marker can be analysed by cytometry,and morphodifferentiation can be observed by confocal microscope.
