摘 要:采用限制性内切核酸酶Hph对山羊肌肉生长抑制素(MSTN)基因的379bp扩增产物进行了PCR-RFLP多态性分析,检测到3种基因型,其酶切位点分别由2种共显性基因控制;对该片段的纯合型分别克隆、测序发现,BB型在第1661位由T→C。上述结果为首次试验证实山羊MSTN内含子2(379bp)区域存在多态性。Myostatin (MSTN) gene was amplified and an uniform fragment of 379 bp was obtained in goat. The 379 bp PCR product was digested with restriction endomuclease Hph Ⅰ and its genetic polymorphism was investigated by PCR-RFLP. Three kinds of genotypes were found in these populations,which were controlled by two alleles. The amplified fragments of AA and BB of the fragment were cloned and sequenced. The result showed that in 379 bp BB genotype had one substitution mutation at 1 661 site(T→C). The result above-mentioned first confirmed that there are polymorphisms in intron 2(379 bp) region of MSTN gene.
