通痹灵对胶原诱导型关节炎大鼠关节破坏的影响

摘　　要：【目的】观察通痹灵对胶原诱导型关节炎(CIA)模型大鼠关节破坏的影响。【方法】采用牛Ⅱ型胶原混合弗氏不完全佐剂于Wistar大鼠尾根部注射复制CIA模型,造模成功后随机分为模型组、通痹灵组(剂量为3.0 g.kg-1.d-1)、甲氨蝶呤组(MTX组,剂量为每周7.6 mg.kg-1),并设正常对照组;各组按设计剂量灌胃相应药物或生理盐水35 d后,采用容积法评价后肢肿胀度,采用乳腺钼靶机进行全身X光照片,比较各组大鼠各关节破坏程度的差异。【结果】通痹灵可显著性减轻CIA模型大鼠的足肿胀度和四肢关节肿胀数,与模型组比较具有显著性差异(P<0.05或P<0.01);模型组大鼠X光片显示各关节骨面受到严重侵蚀,关节间隙变窄,通痹灵可显著性抑制CIA大鼠的关节破坏程度(P<0.05或P<0.01),其作用与阳性对照药MTX相仿(P>0.05)。【结论】通痹灵治疗类风湿性关节炎的作用可能与其抑制关节破坏程度,减轻关节炎症有关。[ Objective] To observe the effect of Tongbiling on articular damage in collagen-induced arthritis （CIA） rats. [ Methods] Wistar rats were injected intradermally with emulsify of Bovine collagen Ⅱ and Freund＇s incomplete adjuvant at the base of the tail to induce CIA rat model. Then the model rats were randomized into model group, Tongbiling group （3.0 g·kg^- 1· d^- 1 ）, and methotrexate （MTX） group （7.6 mg·kg^-1 per weck）. Meanwhile, a normal control group was set up. Except the normal group received saline, the rots in other groups received the corresponding drugs according to the experimental design for 35 days. Hind limb swelling in rats was evaluated by Cubic method. Articular damage in different groups was compared by whole body mammography X-ray radiograph. [Results ] Tongbiling markedly reduced the swelling degree and the number of swollen joints, the difference being significant as compared with the model group （ P 〈0.05 or P 〈0.01）. The radiograms showed that the joints were destructed severely and the joint space became narrow in the model group, and Tongbiling relieved the above joint destruction （ P 〈 0.05 or P 〈 0.01 ）, having similar effect with MTX （ P 〉 0.05 ）. [ Conclusion ] The possible therapeutic mechanism of Tongbiling for rheumatoid arthritis is associated with the inhibition of articular damage and with the reduction of articular inflammation.
