摘  要:目的:探讨一种新型HLA衍生肽(RDP1258)对人外周血单核细胞增殖的免疫抑制作用及其机制。方法:人工固相合成RDP1258,用3H-TdR掺入法观察其在体外对人外周单核细胞增殖反应的影响,以酶化学法观察其对淋巴细胞血红素氧合酶-1(Heme oxygenase-1,HO-1)活性的影响。结果:RDP1258可显著抑制淋巴细胞转化及MLR的增殖反应;该肽体外降低HO-1活性呈现出剂量依赖性。结论:RDP1258能够显著抑制人外周单核细胞经丝裂原或同种抗原刺激引起的增殖反应,这种作用可能是通过影响HO-1活性而达到的。Objective: To explore the immunosuppression of a novel HI.A-derived peptide, RDP1258, on Human peripheral blood mononuclear cells proliferation and its mechanisms. Methods: Peptide RDP1258 was chemically synthesized. The effects of the peptide on alloreactive cytotoxic activities of Human peripheral blood mononuclear cells were observed using 3H-TdR incorporation method. The HO-1 activity was analyzed by the enzyme chemistry method. Results:The results showed that the synthetic HLA-derived peptide could obviously inhibit the proliferation of Human peripheral blood mononuclear cells and inhibit HO activity in a dose-dependent manner in vitro. Conclusion: Heine oxygenase-1 might participate in the RDP1258 inhibiting the proliferation of Human peripheral blcod mononuclear cells induced by mitogen and isoantigen.
