大蒜超氧化物歧化酶活性测定及某些性质研究

摘　　要：采用肾上腺素自氧化法对蒜头ＳＯＤ进行活力测定及某些性质研究。结果表明：蒜头粗提取液富含ＳＯＤ，其比活力为４．６０２×１０４ｕ／ｇ蛋白质；它在３０～６５℃及ｐＨ５～ｐＨ９时活力较强；用ＰＡＧＥ分离出两条酶带；能被高浓度的ＫＣＮ、Ｈ２Ｏ２完全抑制，但对氯仿－乙醇液不敏感，说明此酶为Ｃｕ、Ｚｎ－ＳＯＤ。By using the method of autoxidation of epinephrine,the activity and some properties of SOD in A.Stativum are determined and studied.The result showed that the crude extract from A.Stativum is rich in SOD.Its specific activity is 46.02×10 3 u/g of protein,and can keep under 60℃,pH8 for 10 mins without loss of its enzyme activity.There are 2 bands in the crude extract by using PAGE, which are sensitive to KCN and H 2O 2,and not sensitive to chloroform ethanol,that indicates the enzymes are Cu、Zn SOD.
