摘 要:利用改进的Franz扩散池考察了油酸(OA)、氮酮(azone)和肉豆蔻酸异丙酯(IPM)3种促渗剂(PE)对左炔诺孕酮经离体鼠皮促渗性能的影响。以离体豚鼠皮肤为透皮屏障,HPLC测定左炔诺孕酮含量。结果表明,3种促渗剂的作用机制主要是通过改变角质层的通透性,降低了药物经皮渗透的阻力,提高了药物在皮肤角质层的扩散系数。质量分数为7%油酸(OA)对左炔诺孕酮经皮渗透的促进效果最佳。The in vitro permeability of levonorgestrel across excised cavy skin treated with the penetration enhancers oleic acid, azone and isopropyl myristate has been studied using modified Franz diffusion cells. The analysis was carried out by means of HPLC. It was found that the action of the three skin penetration enhancers leads to the a reduction in the barrier function of stratum cornea, thus reducing the resistance to drug transport through the skin and increasing the diffusion coefficient of levonorgestrel. It was found that addition of 7% of oleic acid gives the maximum increase in penetration of levonorgestrel across cavy skin.
