摘 要:目的探讨多房棘球绦虫重组BCG-Em14-3-3疫苗免疫和Em原头节攻击后小鼠脾CD4+和CD8+T淋巴细胞亚群数量和比值的变化。方法将疫苗采用皮下注射和鼻腔内接种分别免疫BALB/c鼠,免疫后8w用多房棘球绦虫原头节进行攻击感染,感染后18w杀鼠取脾,分离脾细胞,流式细胞仪检测脾CD4+和CD8+T淋巴细胞亚群的百分比,同时设有空载体、BCG和PBS对照。结果疫苗接种组的脾CD4+T细胞亚群显著增加,CD8+T细胞亚群无明显变化,CD4+/CD8+亚群比值明显升高;鼻腔内接种组的CD4+和CD4+/CD8+亚群比值显著高于皮下注射组。结论CD4+T细胞亚群在多房棘球绦虫重组BCG-Em14-3-3疫苗诱导的小鼠抗Em原头节攻击感染的保护性免疫机制中起关键作用,疫苗鼻腔内接种是一种较好的免疫途径。In order to investigate the changes of splenocytes subsets in mice immunized with recombinant BCG-Em14-3-3 vaccine of Echinococcus multilocularis (Era) and challenged with Em protoscoleces, BALB/c mice were immunized subcutaneously and intranasally with recombinant vaccine and challenged with Em protoscoleces 8 weeks later. Then the animals were sacrificed 18 weeks later and spleens were taken to separate spenocytes, in which the percentages of CD4^+ and CD4^+ T cell subsets were determined by flow cytometery. Blank, BCG and PBS were used as controls. It was found that the number of CD4^+ T cell subset in the immunized group of mice increased obviously, but there was no significant change in the number of CD8^+ T cell subset, resulting in a definite increase of the CD4/CD8 ratio. The CD4/CD8 ratio in the intranasal immunization group of mice was significantly higher than that in the subcutaneous immunization group of mice. It is thus concluded that CD4^+ T cell subset may play an important role in the protection of mice induced by recombinant BCG-Em14-3-3 vaccine and the intranasal route appears to be a better way for immunization.
