摘  要:为了理解黄瓜耐热性或热伤害产生的分子机理,以耐热品种‘西双版纳黄瓜’和热敏品种‘二早子’为供试材料,对幼苗进行38℃热激处理2h。采用cDNA-AFLP技术比较热激前后基因表达差异,分离黄瓜热激响应基因,并利用RT-PCR技术检测分离基因的表达。结果表明:从‘二早子’中分离到chrr182(cucumber heat response related)特异片段,经比对发现该片段仅与转录修复偶联因子(transcription repair coupling factor-TRCF)同源;该基因的表达在38℃下被诱导增强;黄瓜幼苗耐热性或热伤害的发生可能与TRCF基因的响应有关。To understand the molecular mechanism of cucumber' s heat tolerance or heat injury, heat-tolerant cuhivar ‘Xishuangbanna’ and heat-sentitive cuhicar‘ Erzaozi' were selected as test materials, and their seedlings were heat treated (38 ℃ ) for 2 hours: The gene was isolated with cDNA-AFLP technique, and the gene expression was detected by RT-PCR technique. A specific fragment of heat response, chrr182 (cucumber heat response-related ), was isolated from the cDNA of ‘ Erzaozi' under heat stress, and the results of blastx in GenBank of NCBI indicated that chrr182 was in homology with transcription-repair coupling factor gene. The expression of chrr182 was enhanced under the inducement at 38 ℃ for 2 hour. The heat tolerance or heat injure of cucumber seedlings could be related to the response of transcription-repair coupling factor gene under heat stress.
