摘  要:从日本引进的西瓜种子隔离种植后,采用生物学、双抗体夹心酶联方法及反转录PCR(RT-PCR)技术对其幼苗样品进行了黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)的检测,结果表明:生物学试验接种的黄瓜健康植株全部发病;双抗体夹心酶联试验结果均为阳性;RT-PCR反应后进行1%琼脂糖凝胶电泳检测,结果出现预期的715 bp特异性扩增产物。3种方法均表明此次从日本引进的西瓜种质已经受到黄瓜绿斑驳花叶病毒的侵染,须做销毁处理。试验结果表明,采用3种方法中的2种对样品进行检测,即可以根据检测结果确诊被检对象是否带有CGMMV病毒。The watermelon seeds imported from Japan were planted isolatedly and their leaves were sampled and detected by bioassay, DAS-ELISA and RT-PCR amplification in the survey for cucumber green mottle mosaic virus (CGMMV). In bioassay, all health plants were infected after they were vaccinated. The result was masculine in DAS-ELISA and one specific band (715bp) was obtained in 1 ; agarose gel electrophoresis of RT-PCR amplification. Positive result for CGMMV was found by bioassay, DAS-ELISA and RT-PCR amplification and it showed that these seeds had been infected by CGMMV. CGMMV could be diagnosed definitely if two of the three tests were masculine.
