摘  要:研究雌二醇对培养小鼠胸腺细胞的影响,发现用1μmol/L、100nmol/L和10nmol/L17β-雌二醇处理胸腺细胞2h能诱导胸腺细胞产生典型的程序性死亡形态学改变和特征性DNA“梯形”结构,并具有剂量依赖性。10μmol/L的它莫西芬可有效抑制1μmol/L17β-雌二醇诱导的胸腺细胞程序性死亡。同时,Staurosporine和放线菌酮亦能抑制雌二醇诱导的胸腺细胞程序性死亡。To investigate whether estradiol might induce apoptosis in mouse thymocytes.The mouse thymocytes were isolated and incubated in culture medium containing 17 β estradiol,17 β estradiol and tamoxifen,or staurosporine,or cycloheximide.DNA fragmentation was visualized by agarose gel electrophoresis.Estradiol elicited typical apoptosis morphological changes and DNA fragmentation in dose dependent manner.The results show that 1 μmol/L,100 nmol/L,10 nmol/L estradiol incubated with thymocytes for 2 h can induce a typical DNA ladder;10 μmol/L tamoxifen could inhibit apoptosis induced by 1 μmol/L estradiol in thymocytes;20 nmol/L staurosporine and 1 mg/L cycloheximide could inhibit the DNA fragmentation.In conclusion,estradiol is able to induce apoptosis in mouse thymocytes and tamoxifen could inhibit it.Protein kinase C and protein synthesis is involved in this event.
