摘  要:为深入阐明严重烧伤早期全身血管通透性增加继而引起早期损害的发生机制,本实验应用培养于微孔滤膜上的小牛肺动脉内皮细胞(EC)单层所构成的选择性通透屏障模型,观察了30%Ⅲ度烧伤大鼠血清对其影响,结果发现,烧伤早期烧伤血清刺激后EC单层对Hanks液或白蛋白液滤过流量、滤过系数均明显增加,而对蛋白渗透压反射系数减少,表明烧伤血清对EC通透性的影响是大鼠烧伤后主要组织脏器水肿的重要因素之一;结合EC活力测定及扫描电镜观察,导致上述改变的原因主要是烧伤血清损伤EC从而造成内皮细胞间裂隙形成或加大,但引起内皮损伤的机制尚需深入阐明。In order to elucidate the mechanism of viscera injury postburn,which mas mainly induced by the increase of capillary permeability,occurring not only in the local burn area but also internal organs,we observed the effects of sera obtained from 30% Ⅲ TBSA burnt rats on calf pulmonary artery endothelial cell monolayer.The cell monolayer was cultured on micropore filter membrane,which could be used to determine filtration volume(Jv),fitration coefficient(Kf)and osmotic reflection coefficient(σ)to protein.Either Hanks balanced salt solution(HBSS)or 5 g/L albumin in HBSS was used to wash the confluent endothelial monolayer at a pressure of 25 cm H 2O.The resuit showed that the exposure of endothelial monolayer to rat burnt sera could significantly enhanced Jv and,Kf values to HBSS or albumin HBSS treatment,and decreased osmotic reflection(σ)to protein.Impaired viability of endothelial cells and morphological changes in endothelial cells were observed by scanning electric microscopy.These results suggest that serum from burnt rat might be an important factor in burn induced increased vascular permeability,and it subsequently leads to visceral edema.The mechanism of endothelial cell injury induced by burn sera needs further study.
